Quantitative Assay Of Uric Acid
Testing the level of uric acid in the blood is a technique that needs to be done periodically to detect the related diseases, the excess or lack of uric acid has bad effects on health.
Definition of Uric Acid
Uric acid is an end product of purine metabolism in the human body
Principle: The colorimetric test uses uricase enzyme to cut uric acid to form allantoin and hydrogen peroxide.
Reaction equation
Uric acid + 2H2O + O2 uricase allantoin + CO2 + H2O2
In the presence of peroxidase, 4 - aminophenazone is oxidized by hydrogen peroxide to form quinone - diimine.
2H202 + H + + TOOSa peroxidase colorant quinone - diimine + 4H202 +
4 - aminophenazone.
The intensity of the concentration of quinone - diimine constitutes proportional to the concentration of uric acid and is determined by measuring the increase in absorbance
Equipment
Biochemical testing machine Biosytem A25
Centrifuges have a maximum speed of 4,000 rpm.
Reagents

Warning
Factors affecting
Jaundice: conjugated bilirubin and unconjugated bilirubin levels> 684 µmol / l or 40 mg / dl.
Hemolysis: when hemoglobin concentration is 1,000 mg / dl or> 621 µmol / l
Lipemia:> 1500 mg / dl. There is poor correlation between turbidity and triglycerides concentration
