Technique For Concentrating Eggs In Faeces

When the amount of parasitic eggs in the stool sample is small, people often use the method of concentrating eggs to detect disease. Using a solution higher than the density of worm eggs, tapeworms, cysts makes them float on the water, the sediment settles to the bottom. The sample is clean, has little sediment, and some eggs do not float but sinks to the bottom.
The technique using saturated brine, also known as the Willis Method
This method is used to find eggs of worms, tapeworms: hookworm eggs, roundworms, hair worms, tapeworm eggs and tapeworms .

..
Do not use this technique to find eggs of tapeworms, schistosomiasis, strongyloid larvae and protozoan active forms.
Rule
In stool testing technique to find parasitic worm eggs, use saturated salt water to dissolve stool
Worm eggs and flukes are lighter than the density of saturated salt, so they float on the surface of the water, stick to glass and are removed for observation under a microscope
Technical process
Put about 5g of stool in a penicillin vial or glass test tube.
Pour into a glass tube some saturated brine, about 1/3 of the vial.

.
Use a special rod to stir up the stool in salt water.
Add saturated brine until the water level is at the top of the glass tube.
Remove the residues that remain on the surface of the water.
Slowly add a few drops of saturated brine to the jar until the surface of the water is curved, then stop (do not let the brine overflow).
Place the lamen over the top of the jar, avoiding the formation of air bubbles between the glass foil and the water

Let stand for about 30 minutes
Lift the lamen upright (the lamen must bring a drop of salt water on the underside) and place it on the slide.
Investigation of the stool sample just made under a microscope.

It should be noted:
If the time is too short, eggs will not have time to float up.

.
If left for too long the eggs will soak into the salt water and sink to the bottom.
The length of time it takes depends on the height of the test tube or bottle. It is best to test the time for the eggs to float with different tubes and bottles.
