Techniques for finding hemoglobin in the stool by Aminopyrine
Principle: When hemoglobin is exposed to Hydrogen Peroxide, Oxygen is released, this Oxygen will affect with Aminopyrine to color.
Patient preparation: A day before the stool blood test, the patient must not eat meat, take iron-containing medicines, brush teeth vigorously causing bleeding.
Chemistry:
10% acetic acid
Hydrogen peroxide
Ethylic alcohol 95o
Aminopyrine
Positive control (1% solution with water-soluble blood)
Negative control (distilled water)
Technical process
Mixing aminopyrine solution: 0

.25g aminopyrine + 5ml of 95o ethylic alcohol
Get some fertilizer into the centrifuge tube. Add 7ml of distilled water and dissolve the stool.
Centrifugal 1000 rpm with time 5 minutes

Pour the supernatant water into a test tube
Slowly put into the test tube the following:
10 drops of acetic acid solution
5ml aminopyrine solution
10 drops of hydrogen peroxide
Take care not to mix substances together
Let stand for 1 minute
Read the results within 5 minutes after adding Hydrogen Peroxide.
Read the result: a positive result will appear red between two layers of solution:
Pale red: positive reaction (+)
Magenta: strong positive reaction (++)
Dark red: very aggressive reaction (+++)
No color: negative reaction (-)

Technical process:
Collect patient's stool in a clean, dry bottle.
Take a stool test stick, at least 3 different places on the stool sample

Put the stool into the vial of diluted stool and mix well
Tear the cover to remove the test strip.
Add 2 drops of stool solution to the test rod into the well with the letter S.
