Microscopic Examination For Helminth Eggs By Kato-katz Technique

Kato-Katz thick smear is a good technique to detect helminth eggs in faeces, especially roundworm, hair, hook. In addition, the detection of schistosomiasis (Schistosoma mansoni) and some other worms and flukes was very effective.
Kato-Katz thick smears are a qualitative and quantitative technique, since the volume of integrals is defined The amount of manure used is so much that it is easier to find worm eggs and flukes than direct stool tests.
Principle: Manure is quantified using a standard measuring hole, placed on a slide and made in a sheet of glassophane paper to absorb glycerin.
Tool
Optical or electronic microscope
Bamboo sticks or dry wood
Stool filter
The stool cover has a measuring hole
Clean glass
Absorbent celophan paper, cut into small pieces
Metal clamp
Absorbent paper towels
Protective gloves
Chemistry
Methylene blue
Glycerin soaked chemicals
Distilled water 100ml
Note: The pieces of celophan paper are soaked in methylene blue solution 24 hours before use

.
Technical process
Place a small stool sample on the newspaper
Push the mesh onto the stool sample so that the stool filters through the mesh and upwards
Place the measuring hole on the glass slide
Using a wooden stick, remove the manure from the top of the net, fill the measuring hole with the stool and remove the excess from the hole.
Take the measuring hole so that the stool in the hole is retained on the slide.
Place a piece of cellophan paper soaked in colored glycerin solution, wipe away the glycerin remaining on the surface of the cellophane paper
Turn the plate upside down on a flat surface, press down gently to spread the stool evenly

Carefully lift the slide by gently sliding to one side, without tearing or separating the cellophane paper
The template has been made to allow 30-60 minutes to spread the stool
Investigation of stool samples just under KHV
Read the results
The stool sample will be clear, un-cleared parasite eggs will be visible on blue background.
This method easily observed worm eggs, high accuracy.

Microscopic Examination For Helminth Eggs By Kato-katz Technique

.
Note:
Hookworm eggs and thin-shelled schist eggs should be fragile if left unattended for a long time. Best to observe after 30 minutes of cellophane paper. This technique uses fresh manure, so be careful when testing.
Evaluate
This technique is simple and accurate and can be conducted in a series of investigations and studies. However, the estimated number of parasitic worms is relatively because worms, flukes do not lay at a regular pace, eggs will not be distributed evenly in the stool
Stool can be thick or liquid, and the amount of manure discharged daily is more or less high
